As far as going to the subfolder that the newdomain is, that should work immediately but what I noticed is that sometimes cpanel adds the .com to the end. If that is what happened to you (you would be able to tell by looking at your folders in your ftp program or cpanel and see what the subfolder is named) then you would have to go to mainsite.com/newdomain.com

I just tried creating an add-on domain myself and it didn't add the .com to the end for the subdomain. I wrote roxy.com in the domain field and it automatically wrote roxy in the subdomain field. Probably a new Cpanel 11 thing. =D
